Senescence-associated metabolic alterations aggravate calcific aortic valve disease.

A new study in European Heart Journal shows that declining NAD+ levels with age contribute to calcific aortic valve disease by disrupting cellular metabolism and promoting inflammation. This research provides mechanistic support for NAD+ supplementation in cardiovascular aging, directly validating the therapeutic rationale for NMN. The findings suggest NAD+ restoration could help prevent or slow progression of this common age-related heart condition that currently lacks effective drug treatments.

Atypical population pharmacokinetics of hydromethylthionine in patients with Alzheimer’s disease explains unexpected phase 3 trial results.

This study examined how methylene blue (hydromethylthionine) is processed in the body when given orally to Alzheimer’s patients versus healthy people. The researchers found that Alzheimer’s patients process the drug differently than expected, which helps explain why previous clinical trials had unexpected results. The drug targets tau protein clumps in the brain that contribute to Alzheimer’s disease and also has other brain benefits.

Development of Freeze-Dried Hyaluronic Acid Sheets for Healing Oral Mucositis: Influence of Hyaluronic Acid Molecular Weight and Nicotinamide Mononucleotide Loading on Healing Efficacy.

Researchers tested different formulations of hyaluronic acid sheets loaded with NMN to treat severe mouth sores that occur during cancer treatment. They found that 800 kDa hyaluronic acid sheets provided the best healing results in animal studies. The NMN-loaded sheets stayed in place well and released the compound steadily, showing promise as a safe treatment for painful mouth ulcers in cancer patients.

β-Nicotinamide mononucleotide preserves muscle strength in septic male mice.

This study found that sepsis patients often develop persistent muscle weakness even after recovery, which is linked to damaged cellular powerhouses (mitochondria) in muscle tissue. Researchers discovered that NMN supplementation helped preserve muscle strength in septic mice by protecting these mitochondria. The treatment worked by supporting a key protein called SIRT3 that helps maintain healthy muscle cell energy production.

Clinical, imaging and blood biomarker outcomes in a Phase 3 clinical trial of tau aggregation inhibitor hydromethylthionine mesylate in mild cognitive impairment and mild to moderate dementia due to Alzheimer’s disease.

This Phase 3 clinical trial tested a methylene blue derivative called HMTM in patients with mild cognitive impairment and early Alzheimer’s disease. The compound works by targeting tau protein tangles in the brain, which are a hallmark of Alzheimer’s. The study measured clinical outcomes, brain imaging changes, and blood markers to assess whether the treatment could slow cognitive decline or improve symptoms.

Neutrophil-Driven Cascade-Targeted Nanocarriers Restore Mitochondrial Homeostasis to Ameliorate Renal Ischemia-Reperfusion Injury.

Researchers developed a targeted drug delivery system that uses NMN (a compound that boosts cellular energy) to treat kidney damage caused by reduced blood flow. In mouse studies, this system successfully delivered NMN directly to injured kidney cells, restored their energy production, and prevented both immediate kidney damage and long-term scarring. The treatment works by boosting NAD+ levels in kidney cells, which helps repair their cellular powerhouses (mitochondria).

Aging Triggers an Intestinal Energy Crisis and HDL3 Deficiency Disrupting Gut-Liver Axis Homeostasis.

This study found that aging causes an energy crisis in the intestines that disrupts production of HDL3, a beneficial cholesterol that protects the liver from inflammation. Supplementing with NMN restored intestinal energy production and HDL3 levels, which helped reduce liver damage and inflammation associated with aging. The research suggests NMN may help maintain healthy gut-liver communication as we age.

Methylene Blue-Enriched Feed Mitigates Acute Nitrite Intoxication in Nile tilapia (Oreochromis niloticus).

This study tested whether methylene blue added to fish feed could protect tilapia from nitrite poisoning, which causes dangerous oxygen transport problems. Fish fed methylene blue for 5 days before nitrite exposure had no deaths and better blood oxygen levels compared to control fish that experienced deaths and organ damage. The methylene blue worked by preventing the conversion of healthy hemoglobin to dysfunctional methemoglobin. This suggests oral methylene blue could be an effective way to protect against oxygen transport disruption.

Modulation of mitochondrial quality by exercise mimetics: A potential strategy for the prevention and treatment of Alzheimer’s disease.

This review examines how exercise mimetic compounds like SLU-PP-332 could help prevent and treat Alzheimer’s disease by improving cellular energy production. When brain cells can’t produce energy properly, it leads to the brain damage seen in Alzheimer’s. These compounds work by activating the same cellular pathways that exercise does, potentially offering benefits for patients who can’t exercise regularly. This suggests exercise mimetics could be a practical intervention for cognitive decline.

Alpha-ketoglutarate accelerates granulocyte-monocyte progenitor differentiation and atherosclerotic plaque inflammation via oxoglutarate receptor 1.

This animal study found that a high-fat diet increases alpha-ketoglutarate levels, which triggers a chain reaction that depletes NAD+ and NMN levels in immune cells. This depletion disrupts cellular energy production and increases inflammation, contributing to atherosclerosis (plaque buildup in arteries). The research suggests that maintaining adequate NAD+ and NMN levels may be important for cardiovascular health and controlling inflammation.
